Cagrilintide (CagriSema): dosage in studies

Cagrilintide has been given to humans under the skin once a week, alone at 0.3-4.5 mg and together with semaglutide at 0.16-4.5 mg [1, 2]A.

In phase 3 trials the dose was 2.4 mg cagrilintide and 2.4 mg semaglutide once a week for 68 weeks, with the dose raised step by step [4, 5]A.

Notes

In the repeated-dose trials the dose was raised step by step over several weeks, and participants were screened [1, 4]A. The kidney and liver studies gave a single dose [8]A.

Nearly all recent trials concern the fixed combination with semaglutide (CagriSema), not cagrilintide alone [4, 5, 6]A.

What points the other way or is uncertain

Adverse events became more common as the dose rose: in the phase 2 trial, 41-63 percent of those on cagrilintide reported gastrointestinal adverse events against 32 percent on placebo [2]A. With the combination in REDEFINE 1, the shares were 79.6 and 39.9 percent [4]A.

Cagrilintide alone lowered HbA1c less than semaglutide, 0.9 against 1.8 percentage points [3]A.
