Sermorelin: dosage in studies
Not a recommendation, not treatment guidance. This page shows only what published human studies used, with sources. This site does not say how much anyone should use of any substance. Fimea warns against using injectable peptides bought online: Fimea warning.
Sermorelin is a 29 amino acid analogue of growth hormone-releasing hormone (GHRH). It has been used into a vein as a test for growth hormone deficiency and under the skin to treat children with growth hormone deficiency [1, 2]B.
In the 1990s two small, short studies in older men used the substance under the name GHRH-(1-29) [3, 4]B.
Doses used in studies
| Study | Participants | Dose | Frequency | Duration | Source and level |
|---|---|---|---|---|---|
| Prakash and Goa 1999 (review, diagnostic test) | Children tested for growth hormone deficiency (review of several studies) | 1 µg/kg into a vein | Single dose | One test | [1]B |
| Thorner 1996 | 110 previously untreated prepubertal children with growth hormone deficiency (open-label study) | 30 µg/kg under the skin | Once a day at bedtime | Up to 1 year | [2]B |
| Corpas 1992, lower dose | 10 healthy older men (9 untreated young men for comparison) | 0.5 mg under the skin | Twice a day | 14 days | [3]B |
| Corpas 1992, higher dose | The same 10 older men, period in random order | 1 mg under the skin | Twice a day | 14 days | [3]B |
| Vittone 1997 | 11 healthy men aged 64-76 with low IGF-I | 2 mg under the skin (self-injected) | Once each evening | 6 weeks | [4]B |
Evidence levels: A = randomised human trial or meta-analysis · B = observational human study or case series · C = animal study · D = cell or laboratory study · E = not peer-reviewed
Units: µg = microgram, written mcg in the calculator. A milligram (mg) is a thousand times larger.
Notes
The children's doses were calculated by body weight, and the treatment dose was for children with growth hormone deficiency [1, 2]B.
The evidence is weak: the studies in older men are from the 1990s, had 10 and 11 participants, and neither had a placebo group [3, 4]B.
What points the other way or is uncertain
2 mg given once each evening did not change IGF-I or body composition over six weeks, and the researchers judged one evening injection less effective than several daily doses [4]B.
In the Corpas study, significant changes in growth hormone and IGF-I were seen only with the higher 1 mg dose, not with 0.5 mg [3]B. According to the review, children's gain in growth rate was smaller than with growth hormone treatment in the studies that gave sermorelin as a continuous infusion or as three doses a day; the once-daily dose had not been compared directly [1]B.
